PAE μSphere™層析基材簡介

μSphere™微球是與Source相同的層析填料基材，是集高流速、高載量、高分辨率(3H)于一體的新一代蛋白純化填料的基礎材料。過去二十多年來，聚合物微球得到了極大發展并成功應用于生物物質分離和純化。PAE開發的聚合物微球，簡寫μSphere™是用聚丙烯酸酯制備的層析介質，傳統的多糖基基材雖生物相容性好，但其骨架剛性低，在較高的壓力下容易出現變形或者孔道的改變，影響純化效率。μSphere™有效解決了瓊脂糖骨架剛性低的問題。

PAE生產的用于生物大分子分離的微球為多孔的，有多種粒徑和孔徑，可達1000A,根須需求接上-OH,-COOH或-NH2 等基團，一邊方便偶聯其它配體或配基。

1. IMAC-μSphere是pae研發的用于金屬螯合層析純化帶有2價金屬離子結合位點的蛋白的填料，適用于所有帶有金屬離子結合位點的蛋白，包括但不限于His標簽蛋白、BSA等。本產品以未加金屬離子的形式提供，用戶可以根據需要選擇適用的金屬離子，如Cu、Ni、Co、Zin等。

2. 尺寸和接口與GE HiTrap系列一致，無死體積。

3. 直連GE、waters、Agilent等公司的HPLC等設備，如lAkta、Prime、Avant等，使用方便，快捷。

This Product is a prepacked column prepared with a column identical to HiTrap from GE, and can be directly linked to Akta and other chromatography equipments, conveniently and fast.
This product is filled with the new generation of polymer mono-dispersed microsphere developed by pae. This new material provides the matrix 3 H features, i.e, High flow rate, High dynamic binding capacity and High performance. All these are very important for chromatography.

This is an affinity purification matrix for His-Tag Fusion proteins and other metal proteins.

本產品的載量遠高于GE同類產品。因此，其洗脫條件可能與GE或其他公司的產品有所不同，可能需要更多的咪唑。方法主要有以下幾點：① 增大洗脫體積；②提高咪唑濃度；③優化洗脫pH。

About HiTalon預裝柱

HiTalon是Ppae為其1ml和5ml預裝柱命名的商標。該商標下的所有預裝柱均可直接與GE、Waters、Agilent等公司的HPLC，如Akta等直連。經轉接可連接BioRad公司的層析系統。

產品描述：

HiTalon預裝柱有1ml和5ml兩種規格。柱體由生物兼容性良好的聚丙烯精工制造，耐壓性高至0.75 Mpa。

兩個規格的接口一致，可以單獨使用，也可以串聯或并聯使用。可以用AKTA工作，也可以與其他層析系統匹配，甚至可以用注射器、蠕動泵或其他方式工作。

每套柱子均包含柱體和若干連接部件和封堵部件，以便與層析系統相連以及使用后密封保存。

IMAC μSphere 6 is composed of highly cross-linked beads modified with a novel chelating ligａnd immobilized to the base matrix. IMAC μSphere 6 is supplied free of metal ions, allowing the user to charge it with the most appropriate metal ion for purification of a target protein.

The coupling chemistry and the novel ligａnd is designed to give minimal ligａnd leakage while providing high capacity.
